ATP6V1C1
ATP6V1C1,全稱為液泡型 ATP 酶 V1 亞基 C1,常用別名 V-ATPase 亞基。它是調控細胞內及細胞器酸化的核心功能蛋白,參與溶酶體功能維持、胞內 pH 穩態平衡、細胞外微環境酸化調節,保障細胞正常代謝、物質轉運與生理活動穩定運行。
ATP6V1C1 異常持續高表達會顯著酸化腫瘤微環境,大幅增強乳腺癌、肺癌、肝癌、胃癌等實體腫瘤細胞侵襲遷移能力,誘導靶向化療耐藥產生,是驅動腫瘤惡性進展的關鍵因子。目前該靶點選擇性小分子抑制劑是抗腫瘤研究熱點,均處于臨床前開發階段,通過抑制異常酸化重塑腫瘤微環境,阻斷腫瘤侵襲轉移,具備廣闊的臨床抗腫瘤應用前景。

IHC image of CSB-PA002399LA01HU diluted at 1:100 and staining in paraffin-embedded human gastric cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

Immunofluorescence staining of MCF-7 cells with CSB-PA002399LA01HU at 1:50, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
