BCAM

BCAM蛋白全稱為基底細(xì)胞黏附分子,也被稱為Lutheran血型抗原或CD239,是一種跨膜糖蛋白。該蛋白主要表達于紅細(xì)胞、上皮細(xì)胞及多種腫瘤細(xì)胞表面,通過與細(xì)胞外基質(zhì)中的層粘連蛋白結(jié)合參與細(xì)胞黏附及遷移過程。研究發(fā)現(xiàn)BCAM在鐮狀細(xì)胞病中異常活躍,其與血管內(nèi)皮細(xì)胞的相互作用加劇了紅細(xì)胞的滯留,導(dǎo)致血管阻塞危象的發(fā)生。在惡性腫瘤領(lǐng)域,BCAM的高表達與卵巢癌、胰腺癌的轉(zhuǎn)移密切相關(guān),可能通過激活特定激酶促進腫瘤細(xì)胞侵襲。近年來針對BCAM的靶向治療逐漸受到關(guān)注,已有研究團隊開發(fā)出阻斷BCAM與層粘連蛋白結(jié)合的單克隆抗體,在動物模型中顯示出抑制腫瘤轉(zhuǎn)移的效果。2022年一項針對鐮狀細(xì)胞病的II期臨床試驗中,BCAM抑制劑聯(lián)合抗凝血療法使患者疼痛危象發(fā)生率降低40%,目前該靶點的雙功能抗體藥物已進入臨床前評估階段。

Immunofluorescence staining of HepG2 cells with CSB-PA341920LA01HU at 1:200, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
