CLDN1
CLDN1，全稱Claudin-1，是一種重要的緊密連接蛋白，在維持細(xì)胞間屏障功能方面發(fā)揮著關(guān)鍵作用。它主要參與形成細(xì)胞間的緊密連接結(jié)構(gòu)，調(diào)節(jié)細(xì)胞旁通透性，以及維持上皮細(xì)胞和內(nèi)皮細(xì)胞層的細(xì)胞間黏附。CLDN1在多種組織中表達(dá)，包括皮膚、肺、胃、腎臟等。其異常表達(dá)與多種疾病的發(fā)生發(fā)展相關(guān)，如魚鱗病、白細(xì)胞空泡、脫發(fā)、硬化性膽管炎綜合征、頭頸鱗狀細(xì)胞癌、肝細(xì)胞癌、結(jié)直腸癌、胃癌、卵巢癌、胰腺癌和甲狀腺癌等。目前，針對(duì)CLDN1的藥物研發(fā)取得了一定的進(jìn)展，例如Alentis Therapeutics開發(fā)的靶向CLDN1的單抗ALE.C04，以及雙特異性抗體ASP1002等，均處于臨床試驗(yàn)階段，顯示出在腫瘤治療和器官纖維化疾病治療方面的潛力。
