DNER
DNER,全稱為Delta/Notch樣表皮生長因子相關受體,常用別名有BET、Bret。它是一種神經組織富集的跨膜蛋白,主要分布在小腦浦肯野細胞與內耳螺旋神經元,參與神經元與膠質細胞的交互調控,對小腦發育、突觸形成及運動協調維持起到關鍵作用。
DNER表達異常與自身免疫性腦炎、小腦共濟失調密切相關,也是抗TR/DNER抗體腦炎的核心靶抗原。同時其高表達會推動肝癌、胃癌、前列腺癌等腫瘤的增殖與侵襲,提示不良預后。目前以靶向DNER的抗體檢測用于神經系統自身免疫病診斷,針對該蛋白的小分子抑制劑與抗體藥物正處于臨床前研發,有望為神經免疫疾病與惡性腫瘤提供新的治療方案。

Immunofluorescence staining of U87 cells with CSB-PA818784NA01HU at 1:50, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
