HAPLN1
HAPLN1,全稱為透明質酸和蛋白聚糖連接蛋白1,常被稱為LINK蛋白。它是細胞外基質中的重要成員,能將透明質酸與蛋白聚糖串聯成穩定的網狀結構,為軟骨、玻璃體等組織提供支撐力,維持它們的彈性和形態完整性。這種連接作用讓軟骨能緩沖關節活動時的壓力,也讓玻璃體保持透明和眼球的正常形狀。
HAPLN1表達或功能出現異常時,可能引發多種疾病。比如骨關節炎患者的軟骨中,它的含量常明顯降低,導致軟骨結構松散、易磨損,進而加重關節疼痛和活動受限;眼部的玻璃體混濁也可能與它的功能變化相關。目前針對它的研究多集中在組織修復領域,比如嘗試通過促進其表達來修復受損軟骨,部分動物實驗已顯示出改善效果,但尚未進入臨床應用階段,未來或為相關疾病提供新的治療思路。

IHC image of CSB-RA179147A0HU diluted at 1:100 and staining in paraffin-embedded human melanoma cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB.
Overlay Peak curve showing HepG2 cells stained with CSB-RA179147A0HU (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100 for 10min. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 35min at 4℃.Control antibody (green line) was Rabbit IgG (1ug/1*106cells) used under the same conditions. Acquisition of >10, 000 events was performed.
