KMO,全稱為犬尿氨酸 3 - 單加氧酶,也叫犬尿氨酸 3 - 羥化酶,是定位于線粒體外膜的黃素蛋白,核心參與色氨酸分解代謝,將犬尿氨酸轉化為 3 - 羥基犬尿氨酸,調控神經活性物質與炎癥介質生成,維持神經、免疫穩態。
KMO功能異常與多種疾病緊密關聯,在亨廷頓病、帕金森病、阿爾茨海默病等神經退行性疾病中,會促使神經毒性物質累積加劇損傷。同時也與精神分裂癥、類風濕關節炎、非酒精性脂肪肝及部分腫瘤相關。目前,RO 61-8048、GSK180 等小分子抑制劑已進入臨床前研究,通過抑制其活性調節代謝平衡,為相關疾病提供新的治療方向。
